چکیده

Mitochondria from rat liver were isolated in 0.25 M sucrose according to Schneider (5) and suspended in 1 ml. of 0.25 M sucrose per gm. of fresh liver. Rabbit liver mitochondria, used only for the experiments in Table I, were prepared by grinding the tissue in 0.25 M sucrose in a Waring blendor at reduced speed (6) and were suspended in 1 ml. of sucrose solution per 2 gm. of liver. The reaction mixture (3 ml.) for measuring oxidation and coupled phosphorylation contained 0.013 M potassium phosphate at pH 7.4, 0.002 M ATP, 0.017 M glucose, 0.003 M magnesium chloride, 0.05 mg. of hexokinase, 0.007 M potassium fluoride, 0.017 M succinate, and mitochondrial suspension

Inhibition of coupling factor B activity by cadmium ion, arsenite-2,3-dimercaptopropanol, and phenylarsine oxide, and preferential reactivation by dithiols.

Coupling factor B activity was measured by the stimulation of the ATP-driven NAD+ reduction by succinate or the 32Pi-ATP exchange activity of Factor B-depleted submitochondrial particles. Half-maximal coupling activity was inhibited by 30 microM cadmium, 5 microM phenylarsine oxide, or 0.3 mM arsenite-2,3-dimercaptopropanol. The inhibition was relieved by slight excess of dithiol but not by a 1...

The uncoupling of photophosphorylation by valinomycin and ammon-ium chloride.

1. Ammonium chloride is a poor uncoupler of photophosphorylation in subchloroplast particles. In the presence of valinomycin the inhibition of photophosphorylation by NH&l is greatly enhanced. 2. The enhancement of the ammonium chloride uncoupling of phosphorylation by valinomycin was less pronounced in chloroplasts than in subchloroplast particles. 3. The light-dependent uptake of ammonium ion...

Cadmium Induced Changes in Metabolic Function of Mitochondrial Isolated from Potato Tissue (Solanum tuberosum L.)

Problem statement: Cadmium is highly toxic at low concentrations, but the mechanism of its toxicity is still not understood particularly at the cellular and subcellular level. Approach: In this study we examined the effects of cadmium on the oxidophosphorylation properties of mitochondria isolated from potatoes. Results: Cadmium strongly disturbed the respiratory metabolism of mitochondria isol...

Uncoupling of energy-linked functions of corn mitochondria by linoleic Acid and monomethyldecenylsuccinic Acid.

Linoleic acid and monomethyldecenylsuccinic acid were tested as uncoupling agents for energy linked functions of corn mitochondria. 2,4-dinitrophenol was used as a standard for comparison. Both compounds uncoupled oxidative phosphorylation, released oligomycin-blocked respiration, and accelerated adenosine triphosphatase. Linoleic acid uncoupled calcium-activated phosphate accumulation and the ...

Effect of rifampicin derivatives on the ion compartmentation of biological membranes.

Based on the experimental results that 3-formyl rifamycin SV acts as an uncoupler in vitro on rat liver mitochondria, the effect of some rifampicin derivatives on the K+ and H+ compartmentation of biological membranes was examined to obtain a chemiosmotic hypothesis for oxidative phosphorylation. The K+ release from mitochondria was remarkably stimulated by 3-formyl rifamycin SV in accordance w...
